[cze][eng] Metoda molekulové dynamiky (MD) umožňuje v reálném čase sledovat vývoj systému složeného z atomů a molekul. Takovým systémem může být např. lipidová dvojvrstva a biomolekula. MD simulace se nejčastěji uplatňuje v návrhu léčiv, kdy laboratorní experimenty s reálným objektem mohou být finančně a časově náročné nebo neproveditelné. Cílem této práce bylo objasnit využití a užitečnost metody MD aplikované na studium lipidových dvojvrstev a ukázat praktické využití této techniky, kterým je studium chování neurosteroidů v lipidových dvojvrstvách. Vhodně navržené a uměle syntetizované neurosteroidy by se mohly uplatnit v léčbě vážných neurologických onemocnění. Součástí této práce jsou i experimentální data získaná MD simulacemi pro vybrané neurosteroidy - pregnanolon glutamát a pregnanolon sulfát. Pomocí MD simulace bylo sledováno chování těchto molekul v modelových membránách a analyzováno. Applications of MD are very common in drug design where the real experimental procedures could be much more financially- and time-consuming or even impossible. The aim of this project is to explain the applications and advantages of MD method in case of studies of lipid membranes, with a special emphasis on a study od neurosteriod behaviour in lipid bilayer. Properly designed and synthesized neurosteroids could be used for the treatment of the serious neurological diseases. This work also included the experimental data obtained by MD simulations for two neurosteroids - pregnanolone glutamate and pregnanolone sulphate. Behaviour of this molecules in model membranes was observed and analyzed by MD simulations.
