[cze][eng] K pochopení fyziologické funkce proteinů je nezbytná znalost jejich prostorového uspořádání, studium konformačních změn a vzájemných interakcí. Chemické zesítění v kombinaci s hmotnostní spektrometrií představuje vhodnou doplňkovou techniku k běžným metodám strukturní biologie (RTG krystalografie, NMR spektroskopie), která umožňuje charakterizovat interakce v komplexech bílkovin za nativních podmínek. Předkládaná práce je zaměřena především na novou síťovací techniku založenou na in vivo inkorporaci analogu methioninu s foto-reaktivní funkční skupinou (foto-Met) do sekvence studovaného proteinu (tzv. proteinová foto-nanosonda). Interakce modelové dimerní molekuly proteinu 14-3-3zeta byla použita pro testování a optimalizaci přípravy proteinové foto-nanosondy a byla tak ověřena využitelnost této techniky k zachycení protein-proteinových interakcí. Byla řešena také interakce membránových hemoproteinů - cytochromu P450 2B4 a cytochromu b5. S využitím chemického zesítění a MS analýzy byla charakterizována vzájemná interakce katalytických domén cytochromů a byly nalezeny dvě možné prostorové orientace cytochromů. Nově zavedená technika síťování pomocí foto-Met umožnila zachytit různé oligomerní stavy katalyticky aktivního membránového komplexu a byl stanoven molární poměr cytochromů v identifikovaných...A comprehensive understanding of physiological role of proteins requires knowledge of their three-dimensional structure, dynamics and protein-protein interactions. Chemical cross-linking in combination with mass spectrometry represents an alternative approach to standard methods for protein structure elucidation (X-ray crystalography, NMR spectroscopy) and enables characterization of interaction interface within protein complexes in their native states. The presented thesis is mainly focused on novel cross-linking methodology based on the in vivo incorporation of methionine analog with photo-reactive functional group (photo-Met) into the sequence of studied protein (so called protein photo-nanoprobe). Interaction between two molecules of 14-3-3zeta protein was used as a model to test and optimize the protein photo-nanoprobe production. The findings confirmed usefulness of this approach for mapping the protein-protein interactions. The photo-initiated cross-linking was used to detect the heterooligomeric membrane structures of cytochromes P450 2B4 and b5 and the molar ratio of cytochromes within individual complexes was assessed.
