Barbora Svobodová Toll-like receptory při bakteriálních infekcích Diplomová práce Univerzita Karlova v Praze, Farmaceutická fakulta v Hradci Králové Odborný pracovník v laboratorních metodách Cíl práce: Stanovení aktivace a exprese toll-like receptorů 2, 4, 5 a 9 pomocí polymerázové řetězové reakce v reálném čase a western blottingem (WB). Zavedení metody WB na pracovišti Mikrobiologického ústavu AV ČR, v.v.i. v Novém Hrádku. Metody: 1) Optimalizace metody WB - extrakce proteinů, měření koncentrace proteinů, elektroforéza, western blotting, imunodetekce proteinů. 2) 4 skupiny gnotobiotických selat: kontrolní bezmikrobní skupina (GF), skupina infikovaná bakterií Salmonella enterica sérovar Typhimurium kmen LT2 (LT2), skupina infikovaná kmenem Escherichia coli O55 (O55) a skupina kolonizovaná Escherichia coli Nissle 1917 (EcN). 3) Real-time PCR - izolace celkové RNA, spektrofotometrické stanovení koncentrace a odhad čistoty RNA, syntéza cDNA, real- time PCR, normalizace a relativizace dat z real-time PCR. Výsledky: Optimalizovali jsme WB detekci TLR5 v tkáňových homogenátech ilea a mezenteriálních lymfatických uzlin. Otestovali jsme protilátky proti prasečím TLR2, 4 a 9. V ileu skupiny EcN byla zvýšena exprese mRNA TLR5 a TLR9. U skupiny O55 nebyl nalezen statisticky významný rozdíl v expresi...Bc. Barbora Svobodová Toll-like receptors in bacterial infections. Diploma thesis Charles University in Prague, Faculty of Pharmacy in Hradec Králové Medical Laboratory Technician Aim: The aim of this diploma thesis was to determine the expression and the activation of the toll-like receptors (TLRs) 2, 4, 5 and 9 in bacterial infections by reverse transcription by real time polymerase chain reaction (RT-qPCR) and by Western blotting (WB). Another aim was to introduce detection of TLRs in a pig by WB in the Institute of Microbiology of the ASCR, v.v.i. in Nový Hrádek. Methods: 1) Optimization of WB - extraction of proteins, protein quantification, electrophoresis, Western blotting, immunodetection of proteins. 2) Four groups of one-week- old gnotobiotic piglets: germfree as a control (GF), piglets infected with Salmonella enterica serovar Typhimurium strain LT2 (LT2), piglets infected with Escherichia coli O55 (O55) and piglets colonized with Escherichia coli Nissle 1917 (EcN). 3) RT-qPCR - purification of a total RNA, spectrophotometric RNA quantification and estimation of purity, synthesis of cDNA, RT- qPCR, normalization and relativization of real-time PCR data.
