Železo je pro většinu organismů jedním z esenciálních prvků jejich metabolismu. Především díky možnosti účastnit se oxido-redukčních reakcí je tento prvek schopen zajišťovat řadu důležitých procesů v buňkách. Na druhé straně je ale známa vysoká toxicita volných atomů železa. Železo je tedy paradoxně jak esenciálním, tak potenciálně toxickým prvkem pro většinu organismů. Pro parazitické organismy je dostatečný příjem železa zásadní vzhledem k jejich vysokým nárokům na množení se ve svém hostiteli. Mnoho studií tedy zkoumá vztah dostupnosti železa na vývoj parazita jako jeden ze zásadních faktorů virulence. V experimentech se využívají chelátory, což jsou chemické látky schopné specificky s vysokou afinitou vázat ionty železa a využívají se především v léčbě nadbytku železa v organismu. Tato práce shrnuje poznatky o účincích nadbytku či nedostatku železa na vývoj parazitických organismů a v této souvislosti také o chelatačních látkách, jejich vlivu na virulenci a využití v terapii infekcí vybraných parazitických protist rodu Plasmodium, Leishmania, Trypanosoma, Trichomonas a Tritrichomonas. Iron is an essential nutrient for metabolism in almost all living organisms. Its importance for many crucial cellular processes originates primarily from the flexibility of available redox potentials. However, the high toxicity of free iron ions is well known. For most organisms, iron is simultaneously and paradoxically essential and toxic. Iron acquisition is crucial for parasitic organisms because it is needed for multiplication in hosts. Many studies have examined the relationship between iron availability and parasite development as a primary factor of virulence. These experiments commonly use chelators, chemical compounds that bind specifically and with high affinity iron ions, and are especially used for iron overload treatment. This thesis summarizes the influence of iron overload or deprivation in the host on the development of parasitic organisms and the impact of chelating agents on the virulence of selected parasitic protists, including the Plasmodium, Leishmania, Trypanosoma, Trichomonas and Tritrichomonas genuses.
