Adipocytokiny uvolňované tukovou tkání hrají důležitou roli v regulaci imunitních a zánětlivých reakcí. Při obezitě dochází k dysregulaci jejich produkce, což je jeden z hlavních faktorů podílejících se na navození chronické celotělesné zánětlivé reakce nízkého stupně, která představuje rizikový faktor pro rozvoj dalších nemocí jako je ateroskleróza nebo diabetes typu 2. Hlavním cílem této práce bylo porovnat sekreci vybraných adipocytokinů (adiponektinu, IL6 a MCP1) izolovaných z tukové tkáně před a po dietní intervenci a to v bazálních podmínkách a během stimolované lipolýzy. Na základě stejného profilu byla následně porovnávána sekrece izoforem adiponektinu. Koncentrace adiponektinu, IL6 a MCP1 byla stanovena pomocí metody ELISA, k určení distribuce izoforem adiponektinu byla použita metoda Western Blot. Dále byla pozornost věnována genové expresi ATF3, ATF4 a HSPA5, faktorů účastnících se stresu ER v průběhu diferenciace adipocytů. Změny genové exprese byly měřeny metodou kvantitativní Real Time PCR. K průkazu správně probíhající diferenciace byly použity geny aP2 a PPARg. V rámci této analýzy byl vytvořen pomocí nepřímé imunofluorescence profil diferenciace adipocytů. Změny sekrece cytokinů (adiponektinu, IL6 a MCP1) v důsledku samotné dietou navozené váhové redukce nebyly prokázány. β... Adipocytokines released by the adipose tissue play an important role in the regulation of immune and inflammatory responses. In case of obesity their production is badly regulated, which is one of the major factors contributing to the onset of a chronic low-grade overall inflammatory response. This response represents a risk factor for the progression of other diseases, such as atherosclerosis or type-2 diabetes. The main goal of this thesis consisted in comparing the secretion of selected adipocytokines (adiponectin, IL6 and MCP1), isolated from the adipose tissue prior to and after a dietary intervention, and this under basal conditions and during stimulated lipolysis. Based on the same profile, the secretion of the isoforms of adiponectin was compared subsequently. The concentration of adiponectin, IL6 and MCP1 was determined by the ELISA method, the Western Blot method was used to determine the distribution of the adiponectin isoforms. The thesis also concentrates on the gene expression of ATF3, ATF4 and HSPA5, factors engaged in the ER stress in the course of the differentiation of adipocytes.
