Imunologický profil u pacientů s roztroušenou sklerózou Abstrakt Roztroušená skleróza je autoimunitní neurodegenerativní onemocnění, které postihuje převážně bílou hmotu mozkovou a míchu. Mechanizmus progrese onemocnění není dosud zcela objasněn. V této studii jsme se zaměřili na srovnání zastoupení imunitních subpopulací a vybraných imunologických znaků mezi pacienty s roztroušenou sklerózou, kteří byli nově diagnostikováni a následně léčeni přípravkem Avonex (IFNβ1a), a zdravými dárci. T buňky se podílejí na patogenezi roztroušené sklerózy a to zejména cytotoxické CD8+ T buňky, které způsobují demyelinizaci axonů. Subpopulace CD161+ Th buněk má potenciál hrát velmi významnou roli v tomto procesu. V naší studii jsme se zaměřili na roli NK buněk jejich fenotyp a funkci v autoimunitní reakci u nově diagnostikovaných pacientů a po následné terapii. Pomocí průtokové cytometrie jsme analyzovali zastoupení NK, NKT, T lymfocytů a monocytů, kde jsme se zaměřili na expresi molekul CD161 a NKG2D na jejich povrchu. Pozorovali jsme zvýšené počty CD161+ buněk v subpopulacích NK CD56bright , NK CD56dim , Th, Tc CD8bright , Tc CD8dim a snížené počty buněk nesoucí znak NKG2D u subpopulací NK CD56bright , NK CD56dim , Th, NKT, Tc CD8bright , Tc CD8dim a monocytů. Immunological profile of multiple sclerosis patients Abstrakt Multiple sclerosis is an autoimmune neurodegenerative disease affecting predominantly the white matter of the CNS and the spinal cord. The mechanism of disease progression is not yet fully understood. In this study we focused on a comparison of selected immunological markers between patients with multiple sclerosis who were naïve newly diagnosed, subsequently treated with Avonex (IFNβ1a) and healthy donors. The T cells (particulary cytotoxic CD8+ T cells) are the major population involved in pathogenesis of MS causing the demyelization of axons. Subpopulation of CD161+ Th cells has a potential to be very important in this process. We focused on the role of NK cells phenotype and function in autoimmune response of patients and their changes during the therapeutic intervention. Using flow cytometry we analyzed the distribution of NK, NKT, T cells and monocytes with special regard to the expression of CD161 and NKG2D molecules on their surface. We observed increase counts of CD161+ cells in subpopulations NK CD56bright , NK CD56dim , Th, Tc CD8bright , Tc CD8dim and decrease counts of NKG2D+ cells in subpopulations NK CD56bright , NK CD56dim , NKT, Th, Tc CD8bright , Tc CD8dim and monocytes.
