[cze][eng] Systém oxygenas se smíšenou funkcí (MFO systém) se významnou rolí podílí na metabolismu endogenních sloučenin i xenobiotik. Tento systém obsahuje cytochrom P450, NADPH:cytochrom P450 reduktasu a také jsou k němu přiřazovány NADH:cytochrom b5 reduktasa a cytochrom b5. Bylo prokázáno, že cytochrom b5 může stimulovat nebo inhibovat reakce katalyzované cytochromy P450, a dokonce může měnit poměr vzniklých metabolitů. Mechanismus působení cytochromu b5 nebyl dosud zcela objasněn. Zjištění protein-proteinových interakcí v MFO systému a určení topologie tohoto systému by mohlo pomoci k objasnění mechanismu působení cytochromu b5. Vhodnou metodou pro identifikaci protein-proteinových interakcí uvnitř membrány je technika kovalentního síťování. Cytochrom b5 obsahuje 3 methioniny, z čehož jsou 2 lokalizovány v C-terminální membránové kotvě. Po záměně těchto dvou methioninů za fotoaktivovatelný analog methioninu (foto-methionin) a následné fotoaktivaci lze určit interakce s cytochromem P450 v membránovém prostředí. Tato práce se zabývá expresí a izolací foto-cytochromu b5 (foto-cyt b5), analogu cytochromu b5 s inkorporovaným foto-methioninem. Podmínky pro inkorporaci foto- methioninu do cytochromu b5 byly optimalizovány a protein byl purifikován s výtěžkem 7,2 mg z 1 l bakteriální suspenze. Izolovaný foto-cyt...The system of mixed function oxygenase (MFO system) participates in significant roles in the metabolism of endogenous compounds and xenobiotics. This system contains cytochrome P450, NADPH:cytochrome P450 reductase, and also there are assigned NADH: cytochrome b5 reductase and cytochrome b5. It was proved that cytochrome b5 can stimulate or inhibit cytochrome P450 (CYP)-dependent reactions and even change the ratio of resulting metabolites. The mechanism of cytochrome b5 action has not been fully elucidated yet. Elucidation of protein-protein interactions in MFO system and determination of topology of this system could explain the mechanism of cyt b5 action. The covalent cross- linking technique is suitable method for identifying protein-protein interactions within the membrane. Cytochrome b5 contains 3 methionines and in 2 cases the methionines are localized in a short hydrophobic C-terminal membrane anchor. Interactions with cytochrome P450 in the membrane environment can be identified by substitution of two methionine for photoactivatable analogue of methionine (photo-methionine) and subsequent photoactivation.
