Cíle dizertační práce: 1. Přes rozsáhlý preklinický i klinický výzkum nebyla dosud možnost preemtivní analgezie definitivně potvrzena ani vyvrácena. V této souvislosti se nám jevily jako zajímavé a podnětné poznatky o analgetickém působení některých antiepileptik za specifických okolností. Cílem našeho výzkumu proto bylo zjistit zda nové a klinicky již úspěšně používané antikonvulzivum levetiracetam má preemptivní nebo i terapeutické účinky v modelu pooperační bolesti u potkanů. 2. Výhodnost některých lékových kombinací byla již prokázána nejen u analgetik, ale i u mnoha dalších lékových skupin. Dříve bylo v naší laboratoři zjištěno, že guaifenesin zvyšuje analgetickou účinnost paracetamolu a kyseliny acetylsalicylové. Druhým cílem našeho výzkumu proto bylo zjistit zda guaifenesin zvyšuje analgetickou účinnost i u nesteroidních antiflogistik (NSA), a to s rozdílnou selektivitou vůči COX-2 (ibuprofen, nimesulid a celecoxib) v modelu viscerální bolesti u myší. Jelikož dříve bylo zjištěno, že guaifenesin zvyšuje absorpci paracetamolu, zjišťovali jsme, zda guaifenesin neovlivní také plazmatické hladiny jednoho z testovaných NSA, a to nimesulidu. 3. Rozsáhlý výzkum svědčí pro to, že aktivace endogenního kanabinoidního systému tlumí bolest. Třetím hlavním cílem našeho výzkumu proto bylo zjistit zda agonista... The aim of our research is was to determine whether new and already clinically successfullyused anticonvulsant levetiracetam has a preemptive or therapeutic effects in the model of postoperative pain in rats. The second objective of our research was to determine whether guaifenesin increases the efficiency even analgesic NSAIDs, with different selectivity for COX-2 (ibuprofen, nimesulide and celecoxib) in the model of visceral pain in mice. As Previously, it was discovered that guaifenesin increases theabsorption of paracetamol, we investigated whether guaifenesin also affect plasma levels of one of the NSAIDs tested, andnimesulide. The third main objective of our research was therefore to determine whether cannabinoid receptor agonist CP-55940 increases the analgesic activity of diclofenac in amodel of visceral pain model in mice and inflammatory pain in rat.
