Tereza Petlánová Konzultant: RNDr. Klára Konečná, Ph.D. Název rigorózní práce: Studium interakce nově syntetizovaných sloučenin s bakteriálním agens. Cíl práce: Cílem této práce bylo zhodnocení antibiotické aktivity látek připravených Katedrou anorganické a organické chemie, Farmaceutické fakulty Univerzity Karlovy v Hradci Králové. Metody: Látky byly testovány pomocí mikrodiluční bujónové metody na osmi kmenech bakterií: Staphylococcus aureus, Staphylococcus aureus methicilin rezistentní, Staphylococcus epidermidis, Enterococcus sp., Escherichia coli, Klebsiella pneumoniae, Klebsiella pneumoniae ESBL pozitivní a Pseudomonas aeruginosa. Výsledky: Látky byly rozděleny do pěti skupin podle společných strukturálních rysů. Nejúčinnější byla skupina salicilanilidových derivátů, kde vykazovalo antibakteriální účinek všech 28 testovaných látek této skupiny. Závěry: Z celkového počtu 73 testovaných látek byl pozorován antibakteriální účinek u 50 z nich. Nejcitlivějšími kmeny byly Straphylococcus aureus, Staphylococcus aureus methicilin rezistentní, Enterococcus sp. a Staphylococcus epidermidis. Naopak žádná z testovaných látek nevykazovala účinnost k modelovému kmenu Pseudomonas aeruginosa. Background: The aim of this thesis was to test antibiotic activity of substances produced by the Department of Inorganic and Organic Chemistry, Faculty of Pharmacy of Charles University in Hradec Králové. Methods: Substances were tested by using microdilution broth method on eight strains of bacteria: Staphylococcus aureus, Staphylococcus aureus methicilin resistant, Staphylococcus epidermidis, Enterococcus sp., Escherichia coli, Klebsiella pneumoniae, Klebsiella pneumoniae ESBL positive and Pseudomonas aeruginosa. Results: Substances were divided into five groups according to associated structural features. The most effective was the group of salicilanilid derivates. In this group, the bacterial effect of all 28 tested substances were shown. Conclusion: The antibacterial effect was demonstrated in 50 from the total number of 73 tested compounds. The most susceptible strains were Staphylococcus aureus, Staphylococcus aureus methicilin resistant, Enterococcus sp. and Staphylococcus epidermidis.
