In this study we have examined arrangement of HP1 proteins and histone modifications related to the centromeric heterochromatin of human and mouse chromosomes. Focal arrangement of HP1beta was characterized by a smaller foci located closer to the nuclear periphery and larger foci associated with the nucleoli of human cells. Heterochromatin of selected centromeres (9cen,14/22cen,Ycen) co localized with foci of HP1beta. Immuno-FISH analyses revealed that centromeres analyzed were H3(K9) dimethylated and absent of H3(K4) dimethylation. V našich experimentech jsme studovali jaderné uspořádání HP1 proteinů (alfa, beta, gama) a vybraných typů modifikací histonů ve vztahu k centromerickému heterochormatinu lidských a myších chromosomů. Uspořádání HP1 proteinů do specifických ohnisek bylo charakteristické tím, že menší ohniska byla lokalizována na periferii interfázních jader, zatímco rozsáhlejší HP1 oblasti byly asociovány s jadérky lidských buněk. Heterochormatin vybraných centromer (9cen, 14/22cen a Ycen) těsně přiléhal k velkým ohniskům HP1 beta proteinu. Immuno-FISH analýzy odhalili, že studované centromery byly H3(K9) dimethylovány, zatímco H3(K4) dimethylace nebyla v těchto oblastech nepřítomna.
